She starred in the Fox comedy Pivoting in 2022. Recently, she was as a character in the Netflix series Murderville, episode-Who Killed Santa.After college, Coupe took classes in improvisational comedy at The Groundlings and ImprovOlympic. In 2003, she toured France as a soldier for an all-female version King Lear. The one-woman show, The Patriots (a sketch show created by her) premiered in November 2005 at the Upright Citizens Brigade Theatre. She was awarded the Breakout Performer Award at HBO U.S. Comedy Arts Festival, Aspen, Colorado. March 2006. Coupe was married to actress and teacher and puppeteer Randall Whittinghill from December 2007 until June 2013.She announced her engagement to Darin Olien, who is the founder of the SuperLife, a lifestyle brand SuperLife, via Instagram on the 27th of November 2014. They were in vacation in New Zealand when they got married on December 24, 2014. They filed for divorce in 2018. Between April 2011 until May 2013 Coupe was a star on ABC's ensemble comedy series Happy Endings, along with Zachary Knighton (Adam Pally), Damon Wayans Jr., Casey Wilson, and Zachary Knighton. Despite receiving critical acclaim as well as a huge fanbase the show was canceled by ABC after the conclusion of the third season on May 3 in 2013. Happy Endings was cancelled. She appeared in a regular role on House of Lies's third season and was also a part of Jay Harrington in Benched, an American comedy on the USA Network that premiered on October 28th on the 28th of October 2014. Then she appeared on the ABC thriller Quantico playing the recurring role of FBI agent Hannah Wyland.
